Melanotan 1

Melanotan 1 is not a peptide that is obtainable in nature, created within the lab and designed to be similar to the endogenous alpha-melanocyte stimulating hormone (α-MSH). Melanotan 1 is 13 amino acids long, identical to α-MSH and only differing in two amino acids from the endogenous peptide hormone. According to the researchers, this structural difference could mean that the synthetic peptide has a greater affinity for the target cells and a longer half-life. This has been further validated by research explaining the two major changes of replacing leucine with norleucine at the fourth position and replaced phenylalanine at position seven with D-phenylalanine.

Two primary melanocortins used in the MSH nature and facilitate α-MSH including MCH1 and MCR3 possibly also bind to at least some degree of eumelanin generation (photoprotective) function. It is believed that this type of eumelanin can protect the tissues from any UV-induced damage. On another note, sebum melanin synthesis has previously been associated with eumelanogenesis in skin cells.

Melanotan shows a high degree of selectivity for MCRs and appears to exert its actions through activation of these MC1, MC3, MC4, and MC5 receptors. Five different melanocortin receptors have been identified: melanocortin 1 receptor (MC1R) through melanocortin 5 receptor (MC5R), with the following postulated functions.
- MC1R - Likely to be expressed in melanocytes (skin cells that may regulate skin/hair colour by promoting the synthesis of melanin)
- MC2R: Known to be present in the adrenal cortex and is thought to be involved in the synthesis of cortisol.
- MC3R: Expressed in the brain and placenta, as well as in various peripheral tissues, MC3R plays an essential role in appetite and energy homeostasis.
- MC4R: Is thought to be present in the central nervous system (CNS) especially in the hypothalamus, and thus it is suggested that MC4R could regulate sexual behavior, penile function and also energy balance in the organism.
- MC5R: Even if it is expressed in many tissues, the precise physiological function of MC5R remains somewhat uncertain; it has been suggested that it may be involved in exocrine processes.

Some researchers imply that Melanotan 1 potentially has affinity enough with the MC1R to stimulate melanin production - resulting in a tan. Importantly, the peptide is thought to have a higher affinity for MC1R than α-MSH and, thus, is predicted to be able to upregulate melanin synthesis and skin pigmentation under conditions of little to no sunlight exposure. More simply, scientists speculate that “Melanotan I indeed causes cAMP production, with MC1R activation and subsequent augmentation of microphthalmia transcription factor (MITF) expression, and thus increases enzymes encoding eumelanic pigment. These hypothesized interactions highlight the intricate nature of Melanotan 1's mode of action and suggest that additional research will be needed to uncover its full range of functions and its potential involvement in specific biological processes.
