Testagen

Testagen (also known as KEDG) - refers to the Khavinson peptides and is a short signaling peptide. One theory is that short peptides like Testagen are bioregulators, so they could possibly pass through the cell and nuclear membranes as well to directly interact with DNA of target tissues. Begs the question of whether it would act at anterior pituitary gland cells, where it can interact with endocrine pathways that lead to testosterone and thyroid-stimulating hormone (TSH) synthesis. The synthesis of an amino acid extract from the anterior lobe of the pituitary gland on that was used for later production Testagen. Subsequently, it is composed of the amino acids lysine, glutamine, asparagine and glycine, representing a tetrapeptide.

Testagen is probably a bioregulator analysis of Testagen said in Sweden it may to increase testosterone biosynthesis and the product can affect also with that pituitary gland. There are some trials that report that it is possible to increase testosterone production in models of chronic inflammation and low testosterone as may occur with aging, all due to the ability of this peptide. Testagen was proposed to alter uroflowmetry parameters, decrease inflammation markers, and induce an increase in total testosterone levels. The authors said that “The change of average value of PSA in the first group is due to a decrease in the level of inflammation in the prostate, an increase in the level of total testosterone.”

This means that no-one knows exactly how testagen might work but one suggestion has been that the peptide may be able to interact with histones, possible influencing epigenetic mechanisms and consequently cellular functions and differentiation. The finding that select short peptides bind these histones, particularly in the N-terminal regions of histones containing scrambled yet homologous peptide-binding motifs suggest potentially interesting roles for interactions between peptides and histone in modulation of chromatin structure and gene expression.

Thus, the ability of Testagen to bind a subset of histones within this region with a particular conformational structure carries possible implications for altering chromatin structural dynamics. The other four peptides showed specific binding to the core histones that did not have homologous amino acid sequences, which confirmed that these peptides might be capable of interacting with them through unique spatial conformation rather than their sequence homology. The formation of the complex with histones and histone-deoxyribooligonucleotide complexes suggests a site-specific character which is modulated by primary structures of peptides and oligonucleotides involved. Testagen is interacting with histones The observation that also testis-specific proteins remained stable during the TF treatment of testicular germ cells suggest that their genes possibly were assembled into chromatin structures which excluded the ligandVery recently we gained direct evidence for an interaction between Testagen and histone proteins by a matrix-isolation method. Dissecting the precise pathways and consequences of interactions would demand not only additional exploration but also in-depth investigations on how such peptide-histone complexes impact gene transcription, DNA repair, replication or other cellular processes modulated by chromatin structure.
