Overview
Sermorelin & Ipamorelin are both synthetic peptides, with Sermorelin consisting of 29 amino acids and Ipamorelin consisting of only 5 amino acids. Sermorelin appears to function as a functional equivalent of natural growth hormone-releasing hormone (GHRH) GHRH is 44 amino acids in length while Sermorelin is truncated to 29 amino acids of this fragment. This short form is also called Growth Hormone Releasing Factor (1-29) or GRF (1-29).
Ipamorelin was the first synthetic GHS reported to have a high selectivity for the growth hormone only causing receptors without significant effects on ACTH and prolactin.
Sermorelin is also referred to as GHRF (1-29) amide, and it was first developed in the early 1980s, but the potential of this fragment has been investigated as far back as the late 1970s. It was postulated that upon introduction, Sermorelin would bind to the GHRH receptors at the pituitary resulting in a secretion of GH.
Both Ipamorelin & Sermorelin are thought to exert their action on the pituitary gland. When bound, these peptides are believed to encourage the pituitary gland to increase production of growth hormone.
Their different method of action is just how they act on the path, on which it exerts its action. Sermorelin seems to work on the GHRH receptors as opposed to Ipamorelin which appears to work via ghrelin pathway. It is, therefore, hypothesized that Sermorelin retains some of the GH-stimulating properties human GHRH, and hence brings similar results as human GHRH to the GH axis in vollunteers. The actions of this hormone are believed to be subsequently mediated by elevations in insulin-like growth factor-1 (IGF-1), which is considered the primary mediator of the anabolic mechanisms resulting from growth hormone production. Most evidence suggests that ipamorelin does not act at GHRH receptors. Recently this artificial pentapeptide to be investigated for use as an growth hormone secretagogue (GHS). It is believed to work in a similar way to the Growth Hormone Releasing Peptides (GHRPs) and may possibly copy the natural rhythm of ghrelin, the hunger hormone. The peptide is believed to act on the ghrelin receptors of the anterior pituitary gland (i.e. Growth Hormone Secretagogue Receptors 1 Alpha, GHS-R1a). This characteristic places Ipamorelin into a distinct class of GH (growth hormone)/Ghrelin secretagogue receptor agonists.
Since the peptides seem to have different mode of action, their combined administration could be utilized resulting in a potentially amplified responses to growth hormone.
Research therefore suggests that whilst Sermorelin may trigger an immediate growth hormone response, Ipamorelin might prolong this process for longer.
